Liposomal NR Delivers Nearly 3× Higher Brain Concentration Than Conventional NR

Liposomal NR Delivers Nearly 3× Higher Brain Concentration Than Conventional NR

Friends working in the NAD⁺ and brain health industries have likely asked themselves the same question:
NR (Nicotinamide Riboside) is widely recognized as one of the most effective NAD⁺ precursors. From healthy aging and combating mental fatigue to supporting cognitive function in middle-aged and older adults, its theoretical benefits appear outstanding.
So why do end users often report results that fall just short of expectations?
The answer lies in drug delivery.
A research team from Huazhong University of Science and Technology published a comprehensive animal study in the Journal of Neurorestoratology, clearly explaining both the inherent limitations of conventional NR and how liposomal delivery addresses these challenges at their root.

PART 01

Why Is Conventional NR So Inefficient at Nourishing the Brain?

The brain is the body's most energy-demanding organ. Every second, it requires sufficient NAD⁺ to generate ATP, sustain neuronal survival, and maintain concentration and memory.
However, nature has equipped the brain with a powerful defense system—the blood-brain barrier (BBB). This barrier actively blocks most hydrophilic molecules from entering brain tissue, protecting the central nervous system from harmful substances.
Unfortunately, free-form nicotinamide riboside chloride (NRC) suffers from three major limitations:

1. Highly Hydrophilic

Its strong hydrophilicity makes it difficult to penetrate the phospholipid-rich blood-brain barrier. As a result, most administered NR remains in the bloodstream, liver, and kidneys, while only a minimal amount actually reaches brain cells.

2. Rapid Clearance

NR has a very short biological half-life. After intravenous administration, it is rapidly metabolized and eliminated, leaving very little active compound in circulation within 24 hours.

3. Extremely High Effective Dose

Earlier animal studies required 300–400 mg/kg intraperitoneal injections before meaningful neuroprotective effects could be observed. Such dosages are impractical for human dietary supplements, both economically and from a safety perspective.
In short, most conventional NR never reaches the brain in therapeutically meaningful concentrations. Without sufficient accumulation in brain tissue, users experience limited benefits, leading to weaker product perception and lower repurchase rates.

PART 02

Scientific Evidence: Liposomal Delivery Dramatically Improves the In Vivo Performance of NR

A 2024 study published in the Journal of Neurorestoratology investigated cerebral ischemia using a uniform intravenous dose of 40 mg/kg, directly comparing free NRC solution with liposome-encapsulated NRC.

1. Plasma Pharmacokinetics: Significantly Greater Systemic Exposure

Compared with free NRC solution at the same intravenous dose (40 mg/kg), liposomal NRC demonstrated:
  • 2.76-fold higher peak plasma concentration (Cmax)
  • 5.32-fold increase in AUC₀–₂₄h, rising from 177.59 to 943.92 mg/L·h
  • A dramatic reduction in drug clearance (209.45 → 43.27 L/h/kg), resulting in substantially prolonged systemic retention
  • No statistically significant differences in mean residence time (MRT) or distribution/elimination half-life between the two groups.

2. Tissue Distribution in Healthy Mice: Enhanced Brain Targeting

The greatest benefit was observed in the brain.
Compared with free NRC, liposomal NRC achieved:
  • 2.93-fold higher brain Cmax
  • 2.68-fold higher brain AUC₀.₂₅–₂₄h
  • 58.8% reduction in brain clearance
These findings indicate that liposomal encapsulation substantially overcomes the limitations imposed by the blood-brain barrier.
Distribution in other organs also changed favorably:
  • Lung AUC increased by 1.51-fold
  • Liver AUC decreased to 61% of the free NRC group
  • Heart AUC decreased to 82% of the free NRC group
Lower non-target accumulation in the liver and heart may also reduce the risk of systemic side effects.

3. Drug Accumulation in Ischemic Brain Tissue

In the middle cerebral artery occlusion (MCAO) model, NRC concentrations in the ischemic hemisphere were significantly higher with liposomal delivery.
At:
  • 1 hour post-ischemia: 2.93-fold higher
  • 6 hours post-ischemia: 2.26-fold higher
Ischemic injury disrupts the blood-brain barrier, and when combined with the membrane-crossing capability of liposomes, NRC preferentially accumulates within the ischemic penumbra surrounding the infarct.
By 12 hours, no significant difference remained between the two groups.

4. Improved Energy Metabolism

Following cerebral ischemia, mitochondrial dysfunction rapidly depletes the brain's essential energy molecules—NAD⁺ and ATP.
Free NRC produced only modest and transient increases in these metabolites.
By contrast, liposomal NRC significantly elevated both NAD⁺ and ATP concentrations at 1, 6, and 12 hours after ischemic injury, indicating sustained mitochondrial recovery and continuous energy support for damaged neurons.

5. Superior Neuroprotection

Reduced Infarct Volume

Compared with the vehicle control:
  • Vehicle: 43.39% infarct volume
  • Free NRC: 35.64% (17.9% reduction)
  • Liposomal NRC: 28.01% (35.4% reduction versus vehicle)
Liposomal NRC provided substantially greater protection than conventional NRC.

Reduced Neuronal Loss and Neuroinflammation

Fluorescent NeuN staining showed significantly fewer lost neurons in the liposomal group.
Quantitative analyses of GFAP and CD11b demonstrated marked suppression of astrocyte and microglial overactivation, indicating reduced post-ischemic neuroinflammation.

Improved Neurological Recovery

Both the Bederson neurological score and the 18-point neurological assessment showed significantly better outcomes in mice receiving liposomal NRC.
Compared with both free NRC and vehicle-treated animals, liposomal NRC-treated mice exhibited:
  • Better motor performance
  • Improved sensory function
  • Better balance
  • Improved reflexes
  • Less post-stroke body weight loss
  • Better overall physical condition

PART 03

What Do These Findings Mean for the Industry?

For companies developing NAD⁺ and brain health ingredients, this study delivers two particularly valuable insights.

First, it confirms that the bottleneck lies in delivery—not the NR molecule itself.

The same NR molecule, when delivered using a liposomal carrier, achieved nearly three times higher brain exposure and significantly stronger protection against cerebral infarction.
In other words, the era of competing solely on raw ingredient quality is reaching its limit. Advanced formulation and delivery technologies are becoming the next key competitive advantage.

Second, it points toward a promising clinical pathway.

Traditionally, most studies investigating NR for brain health have focused on oral dietary supplementation, where therapeutic effects develop gradually over time.
This study, however, demonstrates the potential of intravenous liposomal NR in the treatment of acute cerebral ischemic injury, providing fresh evidence that NR may evolve from a nutritional ingredient into a clinically relevant therapeutic agent.
While the direction is becoming increasingly clear, the real challenge for brand owners and formulation engineers is only beginning.
The true barrier to successful liposomal technology is not the concept itself, but the engineering behind it.
Phospholipid composition, encapsulation efficiency, particle size stability, batch-to-batch consistency, and large-scale manufacturing costs all determine whether impressive laboratory results can be translated into reliable commercial products.
Simply put, the companies that first achieve stable, large-scale production of liposomal NAD⁺ precursors that can be seamlessly incorporated into commercial formulations will be best positioned to capture the next wave of innovation in advanced delivery technologies.
